Predatory bacterial crowdsourcing: Scientists ID simple formula that allows bacteria to engulf food in waves

ScienceDaily (Sep. 27, 2012) ? Move forward. High-five your neighbor. Turn around. Repeat.

That's the winning formula of one of the world's smallest predators, the soil bacteria Myxococcus xanthus, and a new study by scientists at Rice University and the University of Texas Health Science Center at Houston (UTHealth) Medical School shows how M. xanthus uses the formula to spread, engulf and devour other bacteria.

The study, featured on the cover of this month's online issue of the journal PLOS Computational Biology, shows how the simple motions of individual bacteria are amplified within colonies of M. xanthus to form millions-strong waves moving outward in unison. The findings answer longstanding questions about how the waves form and the competitive edge they provide M. xanthus.

"When the cells at the edge of the colony are moving outward, they are unlikely to encounter another M. xanthus cell, so they keep moving forward," said lead author Oleg Igoshin, assistant professor of bioengineering at Rice. "When they are traveling the other way, back toward the rest of the colony, they are likely to encounter other cells of their kind, and when they pass beside one of these and touch, they get the signal to turn around."

Igoshin said the net effect is that the cells "spend more time moving outward than inward, and as a result, they spread faster."

M. xanthus is an oft-studied model organism in biology but, Igoshin said, it is one of the few well-studied organisms that lends itself to the study of systems biology, a rapidly growing field of life sciences that aims to model and discover emergent phenomena -- like the rippling waves of M. xanthus colonies -- that have a basis in genetics but only become apparent when cells cooperate.

"Most of the model bacteria that biologists selected for study in the lab were chosen because they were very good at growing on their own in a test tube and not sticking to the wall or to one another," Igoshin said. "When we were choosing model organisms, we lost a lot of the social properties that systems biologists like to study. M. xanthus is different in that people chose to study it because it grew into cool patterns and structures arising from cooperative behavior."

As a computational biologist, Igoshin specializes in creating mathematical models that accurately describe the behavior of living systems. Such models are useful for understanding the cellular and even genetic basis of emergent phenomena.

In the case of M. xanthus waves, Igoshin and Rice graduate student Haiyang Zhang and postdoctoral fellow Peng Shi created an agent-based model, a computer program that simulated the actions and interactions of individual cells to examine how they collectively produced M. xanthus waves.

The model showed that just three ingredients were needed to generate the rippling behavior:

  • When two cells moving toward one another have side-to-side contact, they exchange a signal that causes one of them to reverse.
  • A time interval after each reversal during which cells cannot reverse again.
  • Physical interactions that cause the cells to align.

To verify the model's accuracy, Igoshin's team partnered with UTHealth's Heidi Kaplan, associate professor of microbiology and molecular genetics. Kaplan and graduate students Zalman Vaksman and Douglas Litwin, both of the University of Texas Graduate School of Biomedical Sciences at Houston, used time-lapse images from microscopes to examine the behavior of M. xanthus waves. The experiments confirmed the relationships between wavelength, reversal time and cell velocity that had been predicted by the model.

"We also found an interesting flip side for the behavior, which was counterintuitive and unexpected," Kaplan said. "The same behavior that causes the waves to spread quickly and to cover newly found prey also allows M. xanthus cells to stay on a patch of food and not drift away until the food is devoured."

The exact biochemical signals that the cells use to sense their prey and signal one another to reverse are still a mystery, but the new study helps to narrow the field. For example, many signals between bacteria do not require physical contact, but the study found that the reversing behavior requires contact between cells.

"If the mechanism for this behavior can be found, it could prove useful for synthetic biologists who are interested in programming touch-induced functions into synthetic organisms," Igoshin said.

The research was supported by the National Science Foundation (NSF). The computer modeling was performed on three NSF-funded Rice supercomputers -- STIC, SUG@R and DAVinCI -- that are jointly managed and operated by Rice's Ken Kennedy Institute for Information Technology and Rice's Department of Information Technology.

France taxes rich and business to slash deficit

PARIS (Reuters) - President Francois Hollande's Socialist government unveiled sharp tax hikes on business and the rich on Friday in a 2013 budget aimed at showing France has the fiscal rigor to remain at the core of the euro zone.

The package will recoup 30 billion euros ($39 billion) for the public purse with a goal of narrowing the deficit to 3.0 percent of national output next year from 4.5 percent this year - France's toughest single belt-tightening in 30 years.

But with record unemployment and a barrage of data pointing to economic stagnation, there are fears the deficit target will slip as France falls short of the modest 0.8 percent economic growth rate on which it is banking for next year.

The budget will also disappoint pro-reform lobbyists by merely freezing France's high public spending rather than daring to attack ministerial budgets as Spain did this week in a bid to avoid the conditions of an international bailout.

"This is a fighting budget to get the country back on the rails," Prime Minister Jean-Marc Ayrault said, adding that the 0.8 percent growth target was "realistic and ambitious".

"It is a budget which aims to bring back confidence and to break this spiral of debt that gets bigger and bigger."

With public debt at a post-war record of 91 percent of the economy, the budget is vital to France's credibility not only among euro zone partners but also in markets which for now are allowing it to borrow at record-low yields under two percent.

The government said the budget was the first in a series of steps to bring its deficit down to 0.3 percent of GDP by 2017 - missing an earlier target of a zero deficit by then.

Of the total 30 billion euros of savings, around 20 billion will come from tax increases on households and companies, with tax increases already approved this year to contribute some 4 billion euros to revenues in 2013. The freeze on spending will contribute around 10 billion euros.

FEARS OF EXODUS

To the dismay of business leaders who fear an exodus of top talent, the government confirmed a temporary 75 percent super-tax rate for earnings over one million euros and a new 45 percent band for revenues over 150,000 euros.

Together, those two measures will bring in around half a billion euros. Higher tax rates on dividends and other investments, plus cuts to existing tax breaks will bring in several billion more.

Business will be hit with measures including a cut in amount of loan interest which is tax-deductible and the cutting of an existing tax break on capital gains from certain share sales - moves worth around four billion and two billion euros each.

Four months after he defeated Nicolas Sarkozy, Hollande's approval ratings are in free-fall as many French feel he has been slow to get to grips with the economic slow-down and unemployment at a 10-year high and rising.

Data on Friday confirmed France posted zero growth in the second quarter, marking nine months of stagnation, as a pickup in business investment and government spending was offset by a worsening trade balance and sluggish consumer expenditure.

Despite a rise in wages, consumers - traditionally the motor of France's growth - increased their savings to 16.4 percent of income from 16.0 percent a year earlier. In another setback, other data showed consumer spending dropped 0.8 percent in August.

"Altogether the data paint the picture of a stagnating economy," said BNP economist Dominique Barbet.

BNP Paribas economist Helene Baudchon said the deficit was likely to come in above 3.0 percent due to the weak growth outlook. "As things stand, achieving a deficit of 3.3 percent would in itself be a remarkable outcome," she said.

School Music Program Loans New Instruments To All 5th Graders ...

Despite schools across Michigan cutting music programs out of the curriculum ? 5th grade students at Warren Consolidated Schools are enjoying the high notes thanks to a new initiative that lets them all learn to play.

It?s just coming into tune, but students like Sydney Marsalais are already learning the scales of success ? and know how rare an opportunity this is.

?Because a lot of those kids would like to really try band and I just feel bad for them, because if they love to actually try to play an instrument then I just feel bad that the don?t,? said Marsalais.

For fourteen years elementary band teacher Catherine Case had to get creative with instrument instruction. But now the sounds of flutes, clarinets, trumpets and drums fill the classroom walls.

Dr. Robert Shaner has trumpeted the program. He?s?the school district?s instruction and technology executive director, and he has lobbied hard to get it off the ground. ?Arts are a 21st century skill. (It) helps kids to be creative and innovative in the classroom and as we know that?s what we want in the 21st century, because the kids are going to have to be really creative to be competitive in the 21st century job market,? said Shaner.

What?s so unique about this program is that each 5th grader receives a new instrument and is offered the opportunity to buy it at the end of the year so they can keep playing for years to come. Case said, ?It?s been proven that students who are in band or a musical group score 27 percent higher over four years.?

But even though Mrs. Case knows how crucial it is to perform ? students like Marsalais just want to play. She said, ?It?s kind of happy that I actually am learning how to play a new instrument that I never played before.?

Out-of-Pocket Costs: High and Rising | Michigan Elder Law Center ...

The last several years of life are often the most expensive due to out-of-pocket expenses that are not covered by Medicare. A new study by the Mount Sinai School of Medicine in Manhattan documents the significant financial burden on individuals and families as result of out-of-pocket costs for older adults at the close of their life.

Published in The Journal of General Internal Medicine, the report analyzed data about 3,209 people who participated in the national Health and Retirement Study and who died between 2002 and 2008. The survey collects information about medical out-of-pocket spending every two years:

  • On average, people with Medicare coverage paid $38,688 for medical care in the last five years of life.
  • There was enormous variation, with 25 percent of participants spending an average $101,791 out-of-pocket for medical services and 25 percent spending an average $5,163 during this period.
  • One-quarter of older adults incurred out-of-pocket medical expenses that exceeded the total value of their assets during this five-year period. Forty-three percent of older adults incurred expenses that exceeded their assets, excluding the value of their homes.

Naturally, this has become a presidential campaign issue, with the Republicans having proposed a voucher system for Medicare that most analysts predict will increase older adults? out-of-pocket costs. According to Robert Moffit, senior fellow at the Heritage Foundation, a conservative research group, all proposals being discussed by the Republicans will require seniors to pay more.

A June 2011 report from the Kaiser Family Foundation found that people with Medicare spent 16 percent of their income on out-of-pocket medical expenses in 2006 (an average $4,241 per Medicare member), up from 12 percent in 1997.

Bill Clinton gets $2 billion in pledges to tackle world's woes

NEW YORK (Reuters) - Healthcare for athletes with mental disabilities, organic know-how for Indian farmers and solar technology for isolated communities were among the pledges made at former U.S. President Bill Clinton's philanthropic summit, which ended on Tuesday.

With its theme of "Designing for Impact" and an emphasis on improving the lives of women and girls in the developing world, heads of states, business leaders and humanitarians at the eighth annual Clinton Global Initiative made 150 new pledges valued at about $2 billion to tackle some of the world's woes.

The three-day summit, scheduled when world leaders are in New York for the annual United Nations General Assembly, also featured appearances from U.S. President Barack Obama and his Republican challenger Mitt Romney, as well as Egypt's new Islamist president, Mohamed Mursi.

"I am convinced that cooperation, not conflict, will define this century," Clinton said.

The meeting is largely a networking opportunity for some of the world's power brokers to exchange ideas and forge partnerships. It also offers a chance for successful companies to highlight projects that demonstrate social responsibility.

On the opening day, Clinton asked Wal-Mart CEO Michael Duke if the company would consider opening a store in the Libyan capital of Tripoli. Duke noted the company has opened stores in some of the world's more troubled cities, but politely declined the suggestion.

The summit was also a family affair. Clinton introduced his wife, Secretary of State Hillary Clinton, who made remarks. And their daughter Chelsea, a board member, moderated a session called "The Case for Optimism in the 21st Century."

The idea for the summit came from Clinton's frustration with attending conferences while he was president that prompted no action. When the initiative began, corporations tended to show up and write checks to fund humanitarian programs. Now many see philanthropy in terms of investment opportunities.

The pledges will impact 22 million people, the Clinton initiative said. Some of the commitments took the form of donations.

In the Democratic Republic of Congo, Suntech Power Holdings and the GlobalECHO Foundation teamed up to install $250,000 worth of solar panels worth $250,000 at a hospital that treats victims of sexual violence.

"When you hear the stories of what they're dealing with, you can't not help them," said Andrew Beebe, Suntech's chief commercial officer.

Drink the Brown ? Part 2 - Fortified Wines Malvasia Portugal ...

Madeira, a fortified wine produced on the tiny Atlantic island of the same name, ages in an equally odd fashion as sherry (see the previous post). The best madeiras end up ageing for years, usually decades, in the attics of lodges in Funchal, cooked by the warm Madeira sun. This method?is called canteiro, as opposed to the less-time consuming estufagem process which involves artificially heating the wine.

Exposing wine to high heat and wide temperature swings for decades at a time is exactly the opposite of how I was taught to?treat fine wine, but it seems to work quite well for madeira. In fact, after suffering through summer after summer in a semi-tropical attic, Madeira becomes quite resilient. After all, what else can you do to the stuff? It can last in the bottle for decades or even centuries.

You?ll see standard madeira blends classified by flavor profile (dry, medium sweet, etc.), but if you?re going to buy some madeira, spend a bit more and go for one with a more specific classification, such as Sercial, Verdelho, Bual and Malmsey, which indicate the predominant grape variety as well as the level of sweetness. Sercial is the driest, and Malmsey the sweetest (you can?read more information about these?styles here and here.) These categories are then further subdivided by age.

At Stella!?in New Orleans, I had a Rare Wine Company ?New York? Malmsey?Special Reserve, produced by Vinhos?Barbeito. ?Malmsey? on a madeira label indicates that it?s made from Malvasia grapes, and the words ?Special Reserve? guarantee that the youngest wine in the blend is at least 10 years old. Although this is among the sweetest styles of madeira, I didn?t have it with dessert. The creative sommelier, Marc J. Doiron,?paired it with some crispy veal sweetbreads with andouille sausage, turnips and egg yolk.

Good heavens, what a marvelous pairing! The madeira smelled rich and woodsy, with some wheat toast in there as well. It tasted predictably sweet and caramelly, but amazing bright acids kicked in on the finish, ensuring that it would be food friendly. It complemented the delicate sweetbreads but stood up to the andouille and turnips as well. Quite the balancing act! I don?t often write ?Wow!? in my notebook, but write it I did.

I don?t currently have any madeira at home, but you can bet it will be on my Christmas list. I could imagine it pairing well with some roast pork with sweet potatoes, or?perhaps turkey with stuffing. Indeed, madeira was quite popular in colonial America, making it a thoroughly appropriate choice for Thanksgiving. Get a bottle now, give it a try, and if you can manage to avoid drinking the whole thing, you can serve the rest to the more adventurous palates at your Thanksgiving table, assuming you have a few. If you don?t, then I say forget hosting dinner and head to Stella! instead. It?s not inexpensive, but my goodness, the food and wine is sheer delight.

Toyota Pushes Hybrid Models, Scales Back Plans for EV

Toyota will roll out 21 hybrid vehicles by the end of 2015, while scaling back plans for widespread sales of its new all-electric compact in a decision that signals the company?s growing confidence in its hybrid strategy and its tepid view of battery-powered vehicles.

Toyota predicted its sales of hybrid models will likely surpass 1 million this year ? nearly double what it sold in 2011 ? and expects to maintain this level of sales through 2015. Despite its confidence in the hybrid vehicles, Toyota acknowledged the need to cut costs to increase profitability and spur sales, the Wall Street Journal reported.

Toyota will release its all-electric eQ on a limited basis in December to local governments and selected users in Japan and the US, a decision that marks the company?s conservative outlook of the market for battery-powered cars. Toyota said a lack of demand for the technology prompted the automaker to drop its plan for widespread sales.

Early demand for EVs has fallen short of projections, according to a Lux Research report that recommended EV manufacturers focus on strategies other than scaling up production to lower Li-ion battery costs and boost flagging sales.

In 2010, Toyota had announced plans to sell several thousand of the all-electric eQ vehicles per year when it originally unveiled the compact EV.

The eQ compact, which is based on the iQ Scion, features a new high-output lithium-ion battery that uses a minimum amount of space, and boasts an improved electric power consumption rate of 104 Wh/km.

Toyota also announced its sedan-type fuel cell vehicle is scheduled for launch around 2015. The vehicle has a power output density of 3 kW/L, more than twice the density of the fuel cell stack currently used in the fuel cell hybrid prototype, but is about half the size and weight. Toyota also developed a high-efficiency boost converter, which increased the voltage, making it possible to reduce the size of the motor and the number of fuel cells.

Maybe it's because I'm young, naive, and easily excitable having yet to be jaded by the pointy elbows of Capitalism, but I think WickedFire could be more. You're thinking,

I took some time out to write this proposal because I have a really big chubby for the idea of community. I've always agreed that once I've established a modest level of success in business, I'd strive to be an impact player at the city and local industry level. It's a win-win. I do believe there are differences between a real world, local community and an online, global forum, but I'll address those if a discussion follows.

Secondly, I've spent too much time around this board for the sake of entertainment and not enough time adding or extracting value. That's going to change, and until I write this community off entirely, I will do my part.

In response to your first question, I'll start by defining what I perceive WickedFire to be. Keep in mind that my first encounter with web design was in 1999 while my first meaningful encounter with SEO was in 2011. WickedFire is a shallow knowledgebase and common for internet marketers and related professionals. And of course, all of the meaningful stuff happens between newly-connected members off-site or offline. But in an industry that is particularly gaurded due to constant warring with search engineers, easily exploitable markets, and a wavering level of ethics, I find it hard to differentiate between the who's who. It was Guerilla who told me, "'Peasant' is just a state of mind." Among the "peasant" pool of our active membership, I see people with at least a basic understanding of marketing (exceptions apply), a commendable drive, and the want to work and learn. To me, that sums up potential. To vets, well, I've heard more than a few remarks regarding the "poor" state of this community in comparison to five years ago. I can't attest to that. But I am a witness to the upside, and I'm one of them. So here's my proposal:

This is not a plea for vets to hire out and mentor a new guy. Instead, leverage the membership here at WickedFire (and no, taking advantage of a situation does not require malicious intent). In my short time here, I can't recall the number of moments I've thought to myself, "Oh snap, that guy is a developer/designer/manufacturer/real estate agent/cock fluffer." Give us - small business founders and freelancers - a way to connect and collaborate beyond STS or the many useful sub-forums currently available.

I suppose this should be directed to Jon, but I'd like to hear what everyone else thinks about this proposal.

I could?ve been like any other man who would have a child and just, you know, live with that woman and continue to just, you know, play the game. I?m tryna do it the right way.
